SimpleDSFviewer

SimpleDSFviewer analyzes differential scanning fluorimetry (DSF) data to characterize protein thermal stability and protein–ligand interactions.

Key Features:

  • Melting Curve Normalization: Automates normalization of melting curves (MC) to ensure consistent interpretation across experiments.
  • Smoothing Algorithms: Applies smoothing techniques to reduce noise and clarify thermal transitions in melting curves.
  • Melting Temperature (Tm) Analysis: Computes melting temperature (Tm) values for assessing protein stability and ligand binding effects.
  • Data Preview: Generates visual previews of analyzed DSF melting curves and derived metrics.
  • Matlab Integration: Implemented in Matlab with integration into the Matlab workspace and compatibility with Matlab Runtime.

Scientific Applications:

  • Protein thermal stability characterization: Uses DSF melting curves and Tm values to assess protein thermal stability.
  • Protein–ligand interaction analysis: Detects ligand-induced Tm shifts to infer binding and ligand effects on stability.
  • High-throughput thermal shift assay analysis: Supports analysis of DSF datasets generated in high-throughput thermal shift assays.
  • Drug discovery, structural biology, and biotechnology: Provides stability and binding metrics applicable to target validation, ligand screening, and protein engineering.

Methodology:

Melting curve normalization, smoothing algorithms, and melting temperature (Tm) calculation are implemented in Matlab, with Matlab workspace integration and compatibility with Matlab Runtime.
