Spot Spine
Spot Spine enables the detection and morphological analysis of dendritic spines in three-dimensional images through an ImageJ/FIJI plugin.
Key Features:
- Spine Detection: Detects local maxima in spine heads for initial spine identification in 3D images.
- Segmentation: Computes intensity distribution around local maxima to segment each spine head.
- Neck Tracing: Traces spine necks from the spine head to the dendrite.
- Morphological Measurements: Retrieves quantitative measurements including spine head volume, spine head surface, and neck length.
- Manual Correction: Provides manual correction capabilities for further control over detection and segmentation results.
- Parameter Optimization: Allows setting of several parameters to optimize detection and segmentation.
Scientific Applications:
- Synaptic Density Analysis: Measures dendritic spine number as a proxy for synaptic connection density.
- Pathology Research: Analyzes altered spine density and morphology observed in several pathologies.
- Learning and Memory Studies: Investigates spine formation and morphological changes that correlate with learning and memory.
- Multi-Method Imaging Analysis: Analyzes images of dendrites obtained with various labeling and imaging methods.
Methodology:
Local maxima are detected in spine heads, and the intensity distribution around the local maximum is computed to perform the segmentation of each spine head. Spine necks are then traced from the spine head to the dendrite.
