SynToxProfiler
SynToxProfiler prioritizes drug combinations by integrating synergy, efficacy, and toxicity data to identify therapeutically optimal pairs.
Key Features:
- Integrated profiling: Integrates synergy, efficacy, and toxicity measurements for each drug combination.
- Selective efficacy scoring: Calculates selective efficacy as efficacy minus toxicity to quantify the therapeutic window.
- Prioritization algorithm: Ranks drug pairs based on combined synergy and selective efficacy metrics to highlight optimal therapeutic candidates.
Scientific Applications:
- Anti-Cancer Drug Combinations: Applied to 20 anti-cancer drug combinations tested on healthy control cells and T-cell prolymphocytic leukemia (T-PLL) patient cells to identify synergistic pairs with higher selective efficacy.
- Anti-Viral Drug Pairs: Applied to 77 anti-viral drug pairs tested in Huh7 liver cell lines with and without Ebola virus infection to identify drug pairs balancing efficacy and toxicity.
Methodology:
Integrates synergy, efficacy, and toxicity data; computes selective efficacy (efficacy minus toxicity); and ranks drug pairs according to combined synergy and selective efficacy metrics.
