tatDB
tatDB catalogs experimentally determined guide–target pairs of transfer RNA-derived fragments (tRFs) to support analysis of tRF-mediated post-transcriptional regulation.
Key Features:
- Extensive Data Repository: Contains nearly 250,000 experimentally determined guide–target pairs involving approximately 23,000 tRF isoforms derived from analyses of chimeric reads in crosslinked Argonaute1–RNA complexes from human HEK293 cells.
- Mechanistic Insights: Provides information on guide–target hybrid secondary structures and identifies binding motifs to support mechanistic interpretation of tRF interactions.
- Hypothesis Generation: Enables generation and testing of hypotheses about tRF function, exemplified by analyses probing roles of tRFs in the SARS-CoV-2 lifecycle.
Scientific Applications:
- Identify and Validate Targets: Retrieve experimentally determined targets of specific tRFs to study their regulatory functions.
- Explore Binding Mechanisms: Analyze guide–target hybrid secondary-structure and binding-motif data to elucidate molecular interactions between tRFs and target transcripts.
- Support Hypothesis Building: Use the comprehensive dataset to generate and test hypotheses about tRF roles in biological processes, including the SARS-CoV-2 lifecycle.
Methodology:
Analysis of chimeric reads from crosslinked Argonaute1–RNA complexes derived from human HEK293 cells to infer guide–target pairs and characterize guide–target hybrid secondary structures and binding motifs.
