Telometric

Telometric analyzes autoradiograph images of agarose gels to measure telomeric fragment sizes and distributions and to correct intensity profiles for multiple telomeric probe hybridizations.

Key Features:

  • Correction for Multiple Hybridizations: Applies corrections to intensity distributions to account for multiple matching telomeric probe hybridizations so that measured intensities better reflect copy number distributions.
  • Statistical Analysis and Visualization: Generates simple statistics and plots for each selected lane to represent telomere size distributions.
  • Nonlinear Migration Consideration: Addresses challenges arising from nonlinear DNA migration on agarose gels when assessing fragment sizes.
  • Focus on Telomeric Repeats: Targets telomeric regions composed of tandem repeats (5′-TTAGGG-3′) for quantitative assessment of telomere lengths.
  • Grayscale Intensity Assumption: Operates under the assumption that grayscale intensity at each image point is proportional to labeled DNA concentration, noting potential limitations for regions <1 kb.
  • Support for Constant-field Agarose Gels: Supports analysis of autoradiographs originating from constant-field agarose gel electrophoresis.

Scientific Applications:

  • Telomere length measurement: Quantifies telomeric fragment size distributions for studies of telomere dynamics and genomic stability.
  • Cellular proliferation and cancer research: Provides quantitative data on telomere length variability relevant to investigations of proliferation and oncogenesis.
  • General DNA distribution analysis: Applies to analyses of labeled DNA distributions on agarose gel autoradiographs and related contexts such as cDNA library characterization.

Methodology:

Scanned autoradiographs of agarose gels hybridized with telomeric probes are imported into NIH Image, where Telometric analyzes grayscale intensity profiles and applies corrections for multiple probe hybridizations and computes per-lane statistics and plots.
