TERA-Seq
TERA-Seq enables direct, true end-to-end sequencing of single, native RNA molecules to characterize 5' and 3' transcript termini and to profile both polyadenylated and non-polyadenylated RNAs.
Key Features:
- End-to-end adapter ligation: Uses uniquely designed adapters ligated to both native 5' and 3' RNA ends to enable identification of terminal regions during sequencing.
- Inclusive transcript capture: Captures both polyadenylated and non-polyadenylated RNA species, expanding detectable RNA populations beyond poly(A)+ transcripts.
- Single-molecule resolution: Operates at single-molecule resolution to reveal per-molecule variation such as poly(A) tail length differences among capped, full-length mRNAs in human cells.
- Capping and processing detection: Identifies capping events downstream of canonical transcriptional start sites in fully spliced and polyadenylated molecules.
- Decay mechanism characterization: Detects cotranslational decay patterns, including mRNAs that degrade from their 5' ends while retaining poly(A) tails.
- Addresses nanopore limitations (Oxford Nanopore Technologies): Enables sequencing of very 5' RNA ends that were previously inaccessible to existing nanopore-based approaches such as those from Oxford Nanopore Technologies.
Scientific Applications:
- RNA biology: Investigating RNA processing, capping, modification, and decay mechanisms at single-molecule resolution.
- Transcriptomics: Generating comprehensive transcriptome profiles that include non-polyadenylated transcripts and terminal-end variation.
- Clinical and diagnostic research: Characterizing RNA species relevant to disease by precise end-to-end profiling of native RNA molecules.
