TFEBexplorer

TFEBexplorer integrates multiple datasets to study genes regulated by the stress-responsive transcription factor EB (TFEB), enabling analysis of gene expression changes and TFEB binding sites across species and cell lines.

Key Features:

  • Gene Query Input: Accepts one gene or a set of genes of interest as input for analysis.
  • Multi-Panel Results Display: Presents results in separate panels reporting experiments or bioinformatic analyses.
  • Expression Change Data: Provides information on expression changes following inducible over-expression, knock-down, and knock-out of TFEB across cell lines and species.
  • Binding Site Information: Reports experimentally validated and bioinformatically predicted TFEB binding sites in the proximal promoter of queried genes.
  • Cross-Species Analysis: Integrates datasets across multiple species for comparative study of TFEB-regulated genes.

Scientific Applications:

  • TFEB Target Gene Identification: Identifies genes modulated by TFEB, the master regulator of lysosomal biogenesis and autophagy.
  • Lysosomal Biogenesis Research: Supports investigation of the Coordinated Lysosomal Expression and Regulation (CLEAR) network controlled by TFEB.
  • Autophagy Pathway Analysis: Facilitates study of gene regulatory mechanisms underlying autophagy.
  • Stress Response Mechanism Studies: Enables analysis of gene expression changes following genetic perturbation of the stress-responsive transcription factor TFEB.
  • Promoter Binding Site Analysis: Examines experimentally validated and predicted TFEB binding sites in proximal promoter regions.

Methodology:

TFEBexplorer integrates multiple datasets following genetic perturbation of TFEB with experimentally validated and bioinformatically predicted binding sites. Gene expression profiles are processed using robust multiarray average (RMA). Chromatin immunoprecipitation followed by sequencing (ChIP-seq) data provides experimentally validated binding sites.
