TheCellVision

TheCellVision hosts a centralized database and analysis platform for high-content fluorescence microscopy images to enable genome-scale, single-cell studies of subcellular morphology and protein localization.

Key Features:

  • Central Repository: Hosts approximately 575,590 fluorescence microscopy images and associated quantitative analysis results from two high-content screening projects on the budding yeast Saccharomyces cerevisiae.
  • Genome-scale single-cell support: Enables genome-scale screens at the single-cell level focusing on subcellular morphology and protein localization.
  • Data types and results: Stores both raw fluorescence microscopy images and derived quantitative analysis results from HCS experiments.
  • Cross-dataset querying: Supports cross-dataset searches and queries to compare subcellular compartment morphology, protein abundance, and localization.
  • Computational tools: Provides computational tools for analysis of HCS datasets and for application to other cell systems, as demonstrated with published mammalian cell images.
  • Yeast ORF-GFP analysis: Supports analysis of yeast ORF-GFP collections under different chemical treatments and mutant backgrounds.
  • Cell cycle and data integration: Enables cell cycle–specific analyses integrating protein localization and abundance with gene expression and translational efficiency metrics.
  • Automated analysis and re-analysis: Facilitates automated analyses and independent re-analyses of existing imaging datasets to support reproducibility and new discovery.

Scientific Applications:

  • Automated protein localization and abundance analysis: Supports high-throughput, automated analysis of protein localization patterns and quantitative protein abundance from fluorescence images.
  • Yeast functional screens: Enables studies of yeast ORF-GFP collections across chemical treatments and mutant backgrounds to link protein localization/abundance to genetic or environmental perturbations.
  • Cell cycle–specific studies: Facilitates analyses that link protein localization and abundance to cell cycle stage and to gene expression and translational efficiency metrics.
  • Cross-species application and validation: Allows application of analytical tools to published mammalian cell imaging datasets for comparative analyses.
  • Data reuse and reproducibility: Enables independent re-analysis of HCS datasets to validate findings and generate new hypotheses.

Methodology:

Provides computational analysis tools for automated processing of HCS datasets, cross-dataset querying, and integration of protein localization and abundance data with gene expression and translational efficiency metrics.
