TRiP

TRiP provides comprehensive RNAi and CRISPR reagent collections for targeted loss- and gain-of-function functional genomics studies in Drosophila.


Key Features:

  • RNAi stock collection: Over 15,000 RNAi fly stocks covering the majority of genes in the Drosophila genome for loss-of-function studies.
  • CRISPR integration: CRISPR-based collections expanded the resource to include both overexpression and knockout reagents.
  • TRiP-OE (overexpression): Uses single guide RNAs (sgRNAs) targeting regions upstream of a gene's transcription start site to enable transcriptional activation.
  • Activation domains: Activation in TRiP-OE is achieved by co-expression of catalytically inactive Cas9 (dCas9) fused to VP64-p65-Rta (VPR) or the Synergistic Activation Mediator (SAM) activator complexes.
  • TRiP-KO (knockout): Uses one or two sgRNAs targeting coding sequence to induce insertions or deletions (indels) for loss-of-function alleles.
  • Tissue targeting: TRiP-KO induces indels in both germline and somatic tissues.
  • CRISPR stock scale: The project has generated over 5,000 CRISPR-OE and CRISPR-KO fly stocks.
