VALERIE
VALERIE visualizes alternative splicing events from single-cell RNA-sequencing experiments and computes percent spliced-in (PSI) values with statistical comparisons across cell groups in R.
Key Features:
- Single-Cell Resolution Visualization: Visualizes alternative splicing events within specified genomic regions across individual cells from single-cell RNA-sequencing data.
- Percent Spliced-In (PSI) Calculation: Computes PSI values for user-defined genomic coordinates associated with alternative splicing events.
- Informative Plot Generation: Generates an ensemble of plots that summarize splicing patterns across cells and cell populations.
- Statistical Analysis Capabilities: Performs statistical tests to compare PSI distributions between user-defined groups of cells.
Scientific Applications:
- Cellular Diversity Studies: Characterizes cell-to-cell heterogeneity in splicing to study cellular diversity within tissues and across developmental stages.
- Disease Research: Detects differential splicing patterns associated with diseases such as cancer.
- Developmental Biology: Maps dynamic changes in alternative splicing during differentiation and development.
Methodology:
In R, VALERIE computes PSI for user-defined genomic coordinates, visualizes splicing events in specified genomic regions at single-cell resolution, generates multiple plot types, and performs statistical tests comparing PSI between cell groups.
