VIRdb
VIRdb integrates curated gene, protein, interaction, and natural-compound data to support molecular analysis and therapeutic target discovery in vitiligo.
Key Features:
- Comprehensive Gene and Protein Data: Contains 129 differentially expressed genes validated through gene ontology and pathway enrichment analyses and identifies 22 genes involved in epithelial cell differentiation regulation.
- Protein Targets and Natural Compounds: Curates 40 protein target molecules and their natural hits identified via virtual screening and includes a curated list of 25,041 natural compounds from the Natural Product Activity and Species Source Database.
- Network Analysis: Provides Protein-Protein Interaction Networks and Gene-Gene Interaction Networks for exploration of interactions among target proteins and differentially expressed genes.
- Drug-target and Systems Biology Integration: Integrates drug-target data with systems biology approaches to enable multi-level analysis of vitiligo molecular mechanisms.
- Analytical Toolkits: Employs gene ontology and pathway enrichment analyses, virtual screening and molecular docking with Schrödinger's GLIDE, network visualization with Cytoscape, and computational analyses on MATLAB server installations.
Scientific Applications:
- Drug discovery and lead identification: Supports identification and prioritization of computationally derived potential drugs and natural leads for vitiligo.
- Target identification and validation: Enables selection and analysis of protein targets implicated in vitiligo pathogenesis.
- Pathogenesis and mechanism studies: Facilitates investigation of differentially expressed genes and epithelial cell differentiation-related genes to study molecular mechanisms of vitiligo.
- Systems-level network analysis: Allows exploration of protein-protein and gene-gene interaction networks to study systems biology of vitiligo.
Methodology:
Curation of differentially expressed genes and protein targets; validation via gene ontology and pathway enrichment analyses; identification of epithelial cell differentiation genes; curation of 25,041 natural compounds from the Natural Product Activity and Species Source Database; virtual screening and molecular docking using Schrödinger's GLIDE; construction and analysis of Protein-Protein Interaction and Gene-Gene Interaction networks with Cytoscape; computational analyses on MATLAB server installations.
