VisualCNA

VisualCNA performs constraint network analysis and visualization to assess biomolecular structure, graph-theory–based rigidity, flexibility, thermostability, and function.

Key Features:

  • PyMOL integration and visualization: Integrates with PyMOL to display CNA results alongside molecular graphics representations, mapping rigidity and flexibility onto structures.
  • Constraint Network Analysis (CNA): Applies graph theory-based rigidity analysis to assess how structural constraints contribute to overall stability and to identify rigid and flexible regions.
  • Integration with FIRST: Interfaces with FIRST (Flexible Structure-Related Thermodynamics) software to aid assessment of protein flexibility and stability.
  • Protein engineering for thermostability: Links rigidity and flexibility analyses to identify structural determinants and potential modifications to enhance thermostability.

Scientific Applications:

  • Thermostability engineering: Identification of structural determinants of protein rigidity to guide design of variants with enhanced thermal stability for industrial or therapeutic applications.
  • Structure–function and flexibility studies: Elucidation of relationships between molecular flexibility, rigidity, and biological function by mapping constraint networks onto structures.

Methodology:

Performs graph theory-based rigidity analysis via CNA to evaluate structural constraints and maps those constraints onto molecular representations; integrates FIRST to assess protein flexibility and stability.
