ICGEC

"ICGEC" (Integrative Comparative Genomic Epigenetic Conservation) is a computational method to quantify the degree of epigenetic conservation between two cell lines. Recognizing the pivotal role of histone post-translational modifications in the epigenetic regulation of gene expression and their association with phenotypic differences across cell types, ICGEC aims to overcome the limitations of existing comparative approaches that focus primarily on the absolute differences in levels of individual histone modifications.

Key Features and Functionalities:

- Quantitative Epigenetic Conservation Scoring: Unlike traditional comparative methods, ICGEC provides a quantitative score reflecting the relative change in the epigenetic context of a gene under two different conditions, alongside a separate score for each histone mark involved.

- Application Across Multiple Histone Modifications: The method has been applied to analyze data involving 16 types of histone modifications, demonstrating its capacity to handle complex epigenomic datasets.

- Insights into Cell Differentiation Processes: By identifying epigenetically dynamic and conserved genes based on ICGEC scores, the tool offers novel insights into the biological processes underlying cell differentiation, surpassing the limitations of traditional expression analysis.

- Identification of Key Histone Marks: The analysis facilitated by ICGEC has pinpointed H3K79me1 and H3K27ac as critical histone marks influencing the differential expression of epigenetically dynamic genes between cell lines, highlighting their importance in cell identity and differentiation.
