cellHTS2
"cellHTS2" addresses the challenges of analyzing data from high-throughput RNAi screening, mainly focusing on reducing false negatives and false positives in gene discovery studies. A meta-analysis of various genome-wide, cell-based Drosophila RNAi screens, including a focused RNAi screen, found that false negatives occur at a significant rate of at least 8%. The software demonstrates that integrating knowledge of the cell transcriptome can clarify ambiguous results, enhancing the reliability of RNAi screening outcomes.
The analysis underscores the variability among RNAi reagents targeting the same gene, often leading to inconsistent results due to false positives and weak or ineffective reagents. CellHTS2 suggests these issues can be mitigated by employing RNAi libraries that include multiple independently-tested reagents per gene. This approach helps minimize false positive results through transcriptome data filtering and reduces false negatives, thereby increasing the accuracy of RNAi screens.
